Diacylglycerol Kinase δ: A pivotal player in diacylglycerol induced insulin resistance?

The recent paper by Chibalin et al. contributes a new mechanism to our understanding of hyperglycemia-induced glucose toxicity. The authors show that hyperglycemia inhibits DGK-δ activity, leading to an intramyocellular accumulation of diacylglycerol which may then cause insulin resistance by modulating insulin signaling via a PKC-mediated inhibition of the IRS/PI3K/Akt pathway.
